Human growth hormone (hGH) consist of 191 amino acids, 22kDa molecular weight is recombinant protein that used for treatment of turner syndrome and short stature. Escherichia coli host was used for the production of non-glycosylated hGH as inclusion body. In this work, hGH gene was cloned in pET 21a vector, then transformed into Rosetta-gami (DE3) strain. Recombinant protein expression was induced by IPTG, after observation gel electrophoresis, high expression in media with Rosetta-gami (DE3) [pET21a-rhGH] strain was detected. The aim of this study was to optimize the ethanol and MgSO4 factors that affecting the production of rhGH in E.coli using RSM design in shake flasks.We investigate the effect of ethanol and MgSO4 concentration on E.coli growth and production of hGH, Central composite design from RSM method was used and results showed that increasing the concentration of MgSO4 in the presence of ethanol have a significant impact on the expression of hGH. Protein expression concentration 1.155 g/l in batch culture was obtained with 4 g/l MgSO4 and 1% ethanol.
