In this study, metabolic flux analysis method was applied for calculation of metabolic flux distribution of mycophenolic
acid (MPA) biosynthesis by Penicillium bervicompactum. An over-determined metabolic network model including 68
biochemical reactions and 70 cell metabolites was developed for MPA biosynthesis in P. bervicompactum. Three
extracellular fluxes (glucose consumption and biomass and MPA production fluxes) in steady-state condition in a
continuous 2-L stirred tank bioreactor were determined. Stoichiometric representations of 68 metabolic reactions, mass
balance equations for all cell metabolites and a 70×68 stoichiometric coefficients matrix were determined as tools for
analysis of the metabolic fluxes. Metabolic flux distribution was calculated by least-square method using MatLab
software. Finally, consistency of the developed metabolic model was approved with calculated extracellular metabolites
fluxes.
