Aim and Background: Synthesis of non-toxic and biocompatible nanoparticles is very important subject in biomedicine and drug delivery as most of the nanomaterials are synthesized using toxic chemical reagents. Green synthesis makes use of plant constituents as reducing and stabilizers agents to synthesize nanoparticles. On the other hand, Selenium is an essential trace mineral that supports many bodily processes and it can help improve physiological properties and medical treatment. In the present study, Melissa officinalis L. extract have been used for synthesis of non-toxic selenium nanoparticles (Se NPs). Methods: In this research, simple and fast procedure was developed for Se nanoparticles synthesis. Melissa officinalis L. leaf was processed to obtain aqueous extract as a biological agent for nanoparticles production. Synthesized nanoparticles were characterized by UV-Vis spectroscopy, DLS, ζ-potential, and SEM. Furthermore, toxicity of the Se NP was evaluated using MTT assay on normal fibroblast cell line after various times of exposure. Results and discussion: The synthesized Se nanoparticles using UV-visible spectrophotometry showed absorbance peak in range of 280-285 nm. DLS and ζ-potential analyses indicated the successful formation of Se NPs with particle size of 68 nm and stable colloidal solution, respectively. SEM images of the Se NPs showed the spherical shape. Moreover, MTT assay for fibroblast normal cell lines exhibited non-toxic and reveal biocompatibility of prepared Se NPs at the treated concentrations. Conclusion: Results of this investigation confirmed this protocol as rapid, simple, and a non-toxic alternative conventional chemical methods. Plant extract mediated synthesized Se NPs are biocompatible, non-toxic and can be used in the medicine and biological application
