Aim and Background: Interaction of drug delivery systems (DDS) with plasma proteins such as human serum albumin (HSA) can dramatically affect stability and DDS efficacy. This study aimed to investigate the interaction of crocin and its nanoniosome with human serum albumin. Methods: UV-vis spectroscopy, fluorescence spectroscopy, and circular dichroism (CD) spectroscopy were used to investigate the structural effects of HSA after interaction with crocin and crocin nanoniosomes. Results and discussion: Based on UV-vis and fluorescence spectrophotometric results, nano-niosomes interacted with HSA with a higher binding constant than crocin. The CD spectrum showed a decrease in the α helix content of HSA, especially after crocin addition compared to the crocin nanoniosome. Conclusion: This study shows that drug delivery carriers such as synthesized nanoniosomes have an essential role in reducing the dosage of drugs and side effects.
