Introduction: due to the impossibility of skin absorption of protein compounds such as human growth hormone (hGH), the research and development departments of the cosmetic industry are facing a big challenge for topical administration of growth hormone. Methods: In this study, nanoliposome was used in different cream bases to facilitate skin absorption hGH . Liposomes were synthesized using thin film hydration method. Then characterization of nanoliposome (size, PDI and zeta potential, morphology and effective encapsulation) was done. Cytotoxicity of nanoliposome containing growth hormone was performed. For the local administration of liposomes, different cream-based formulations were used and the stability of nanoliposomes in the formulations was evaluated. Then skin absorption of free growth hormone, growth hormone loaded in liposome and liposome loaded with growth hormone in the cream base was measured. In the end in vivo, histological studies were performed on mouse skin. Results: size, PDI and zeta potential of liposome-hGH are 100 nm, 0.2 and -36.4 mv respectively. The effective loading rate of nanoliposome is 40%. No significant cytotoxicity was observed for nanoliposome-hGH compared to the control group (non-treatment). Skin absorption loaded hGH in nanoliposome increased significantly compared to free hGH. For the local administration of nanoliposome-hGH, a cream formulation that provides conditions for the stability of nanoliposomes was selected. In histological studies, it was found that the thickness of the epidermis increased from 70 nm to 170 nm Conclusion: Our findings showed that it is possible to prepare a cream-based formulation that can facilitate the local administration of growth hormone while maintaining the stability of the liposome containing hGH. Nanoliposome containing hGH significantly increased skin absorption of hGH and subsequently the thickness of skin epidermis.
