Introduction: One of the main challenges in cancer treatment is the side effects of current chemotherapy agents, which have limited the use of these compounds. Therefore, providing novel chemotherapy agents with less side effects and more efficiency is the need of the hour for cancer treatment. Meanwhile, natural products play a significant role in the development of chemotherapy agents. Studies have reported that natural products have promising therapeutic potential with minimal side effects. They can also be effective in reducing adverse effects caused by chemotherapy. Sesquiterpene lactones are a group of natural products that have attracted the attention of many researchers in the field of cancer. These compounds are one of the largest groups of natural products. There are more than 5000 types of compounds in plants, especially the Asteraceae family. Accumulating evidence showed that sesquiterpene lactones have anti-tumor effects in various types of cancers in vitro and in vivo. Therefore, these compounds can be a potential and promising source in the development of chemotherapy agents. Methods: We performed a comprehensive search of the scientific data published on the anticancer activity of sesquiterpene lactones from various databases, including Web of Science, Google Scholar PubMed, and Scopus. Results: The results indicated that sesquiterpene lactones affect different signaling pathways involved in cell growth and proliferation, cell cycle, apoptosis, metastasis, and angiogenesis of cancer cells. Also, accumulating evidence revealed that these compounds can inhibit many human tumors in vitro and in vivo. Conclusion: Given the different antitumor impacts of sesquiterpene lactones in different tumors, these compounds can be a promising potential source in the development of novel chemotherapy agents.
