Toxicity assessments of polymeric nanoparticles in biomedicine

Polymeric nanoparticles more specifically micelles have shown great therapeutic potential in biomedicine and drug delivery but their toxicity has impaired their clinical application. In this context, biocompatibility, hemocompatibility, cytotoxicity, and acute toxicity assessments were conducted to reveal the potential toxicity of PEGylated fatty acid (linoleic acid). These amphiphilic drug vehicles can undergo self-assembly in an aqueous solution to form micelles. The results indicated that developed polymeric micelles did not show any significant toxicity against Artemia Salina which highlights the safety of fabricated drug carriers (100% viability). Moreover, hemotoxic effects (less than 2% hemolysis) caused by these nanoparticles were negligible and their hemocompatibility is thus underlined. No death was recorded by oral administration of PEGylated linoleic acid (acute toxicity) in treated mice even at a high concentration of 5000 mg/Kg and the utility of developed PEGylated linoleic acid nanoparticles as a nontoxic drug carrier is thus proved. Furthermore, the viability of HEK 293 (normal cells) treated with fabricated polymeric micelles was approximately 100 % which demonstrated that these drug vehicles had no toxic effects on normal cells. Altogether, these findings confirm the usefulness of PEGylated fatty acid as a propitious and nontoxic drug carrier for a broad range of chemotherapeutics.
