One of the most critical challenges of bone tissue engineering is overcoming hypoxia conditions, in which oxygen-generating biomaterials (OGBs) play an essential role. In this research, magnesium peroxide was synthesized as one of the OGBs by adding hydrogen peroxide to the reaction with flow rates of 0.1248, 2.62, and 5.24 ml and then loaded into PLGA microspheres. Microscopic images show an increase in the diameter of magnesium peroxide nanoparticles (from 150 to 320 nm) with an increasing flow rate. Also, the diameter of PLGA microspheres increased from 9.72 to 29.73 µm by loading magnesium peroxide. Oxygen release of the samples was stable in the range of 7.2 to 8 mg/l, and magnesium release was 15 to 30 mg/l during 14 days. The cell viability of PLGA-MS and PLGA-MS+MgO2 during seven days was equal to 87.2 and 90.82%, respectively, indicating a suitable environment for cell growth and differentiation for bone tissue engineering.
