چكيده فارسي :
The tropane alkaloids hyoscyamine and scopolamine are widely used as anticholinergic drugs that
act on the parasympathetic nervous system. The only source of the tropane alkaloids are
Solanaceae plants such as Atropa, Datura, Duboisia, Hyoscyamus and Scopolia. In the
biosynthetic pathway leading to the scopolamine, the enzyme hyoscyamine 6-β-hydroxylase
(H6H; EC1.14.11.11) catalyses two consecutive steps; the hydroxylation of hyoscyamine to 6-ßhydroxyhyoscyamine,
and the epoxidation of 6-ß-hydroxyhyoscyamine to scopolamine. In this
study, the Plantlets of Atropa belladonna werecultured in a modified MS medium containing 0.1
mM IAA and 1mM BA, under different concentrations of Cr (0.2, 0.5 and 1 mM). After a month,
roots were appeared. Then, total cellular RNA of the aerial parts (including stems and leaves) was
extracted. cDNA of H6H gene was synthesized by specific primers. Gene expression was
monitored by semi quantitative RT-PCR.>
