Western and Dot Blot Technique without the need for Dark Rooms and its Equipment

وسترن بلات یكی از متدهای ایمنواسی میباشد كه برای آشكار سازی پروتئین در بافتها و مایعات مختلف كاربرد دارد. این روش یك روش تركیبی از الكتروفورز و انتقال پروتئین ها به یك فاز جامد می باشد. این روش در مقایسه با تست الیزا اختصاصی تر است ولی از حساسیت كمتری برخوردار است و چون آزمایشی نسبتاً گران محسوب می شود. به عنوان اولین آزمایش انجام نمی گیرد و بیشتر در تأیید نتایج مثبت شده تست الیزا بكار می رود. یكی از عوامل گرانی این آزمایش نیاز به اتاق تاریك و تجهیزات آن می باشد. استفاده از محلول TMB (كه اصولا در تكنیك ELISA كاربرد دارد) جهت آغشته نمودن غشاء نیتروسلولزیی است كه قبلا باندها از ژل SDS-PAGE به این غشاء منتقل شده و كلیه مراحل وسترن بلات اعم از آنتی بادی های اولیه ، ثانویه و آشكار ساز لومینوسانسی، روی آن ریخته شده است می تواند بخوبی باندها را ظاهر كند. با عكاسی از این غشاء دیگر نیازی به مراحل اتاق تاریك نمی باشد.

Western blotting is a method to detect proteins in tissues and fluids. This method is a combination of electrophoresis and transfer of proteins to a solid phase. This method is compared with the ELISA test, is more specific but less sensitive and because the test is relatively expensive , as the first test was not carried out and further confirm the positive results of the ELISA test is used One of the expensive reasons is these tests require dark room and its equipment. The using of TMB solution which principally is used in ELISA technique for imbruing the protein transferred nitrocellulose membrane which all primary and secondary antibody and luminescence detector were pour on it can appear the bands well. By taking a camera picture the dark room is not necessary
