Abstract :
In this study factors affecting the germination, protocorm body formation and plantlet regenerationof Orchis sancta were investigated. Due to intense contamination occurring fallowing in vitroconditioning of surface sterilized O. sancta seeds, a preparatory step was devised and this stepis unique in that, to our knowledge such step has not been formulated yet in any other previouswork. Fallowing this preparatory step a surface sterilization method practiced, aiming minimumcontamination and maximum viability during in vitro culture. For maintaining this aim and ascontinuing step of surface sterilization, germination media was seeded with 0.02% (w/v) AgNO3.The basic culture media composition included simple inorganic salts that are being used readilyfor orchid culture in vitro. During germination and protocorm body formation this basic culturemedia composition was supplemented with various types and concentrations of nitrogen sources.Germination and protocorm formation scores were arbitrarily taken at 10, 30 and 50 days. According to the results, in this work, O. sancta protocorm formation was maintained at highest rate (47%) in tryptone containing media. These protocorms later on were transferred to plantlet regeneration media that is fortified with two organic substrates. Approximately 1 month later, O. sancta protocorms transferred to plantlet regeneration media were converted to 5-7 cm long small plantlets where conversion rate was as high as 96%.
