Comparison of two different real time PCR tests for hepatitis B virus quantification: COBAS AmpliPrep/COBAS TaqMan AND ARTHUS QS-RGQ KIT
Abstract :
Aim: The aim of the study is to compare two real time PCR tests, the Cobas AmpliPrep/COBAS TaqMan HBV test v.2 (Roche Molecular Systems-USA) and the Arthus HBV QS-RGQ Kit (Qiagen, Germany).Materials and Methods: Eighteen clinical plasma samples and eight QCMD samples were included. Nucleic acid extractions and amplifications were performed according to the manufacturer’s instructions. Samples were tested in duplicate for intra-assay evaluation of Arthus HBV Kit and once again in a different run for inter-assay evaluation. The log10 IU/ml values of the results were evaluated with regression analysis.Results: The coefficient of regression analysis for the COBAS TaqMan HBV test v.2 versus the average of the Arthus HBV Kit results was 0.91 (R2=0.91) and the logarithmic difference was 0.08-1.26. The log differences for inter-assay and intra-assay evaluations were 0.07-0.41 and 0.02-0.19 respectively. The coefficient of the regression analysis was 0.99 (R2=0.99) for both inter-assay and intra-assay evaluations. The log differences for the Arthus HBV Kit QCMD results versus expected values were 0.60-0.89 and the coefficient of regression analysis was 0.99 (R2 =0.99). The log differences for inter-assay and intra-assay evaluations with QCMD samples were 0.04-0.1 and 0.01-0.07 respectively. The coefficient of the regression analysis was 0.99 (R2=0.99) with QCMD samples for both inter-assay and intra-assay evaluations.Conclusion: The Arthus HBV Kit produced accurate and reproducible results. The results of the COBAS TaqMan HBV test v.2 and Arthus HBV Kit were comparable. All log differences were within 1 log except for one clinical sample. In spite of comparable results, the use of the same test during patient follow-up is still important.
