The Effects of Losartan on Immunoreactivity of Irisin on Brain Tissue of Experimental Diabetic Rats
Abstract :
Objective: Diabetes Mellitus (DM) is a hyperglycemic metabolic disorder that alters the energy metabolism. It was also observed that DM causes pathological changes on the central nervous system in both humans and experimental animal models. In this study; we aimed to investigate the effects of losartan (LS) on immunoreactivity of irisin on brain tissue of experimental diabetic rats.Material and Methods: In this study; 18 adult male Wistar albino rats were used. Experimental rats were divided into 3 groups consisting 6 rats in each group. In the control group; the rats didn t receive any drugs during the 6 weeks of study period. In the DM group; 50 mg/kg single dose streptozotocin (STZ) was administered intraperitoneally (i.p). In the DM+LS group; 50 mg/kg single dose STZ was administered i.p and 10 mg/kg/day LS was started orally after the confirmation of DM. At the end of the experiment, all of rats were euthanized by cervical decapitation and brain tissues were removed and fixed. The brain tissues were processed to determine immunohistochemical localization of irisin by using avidin-biotin-peroxidase technique. Histoscore was created according to the diffusiveness and severity of immunoreactivity in assessment of immunohistochemical staining. Results: There was a statistically significant decrease in the DM group in terms of irisin immunoreactivity compared to the control group. Irisin immunoreactivity was significantly increased in DM+LS group compared to DM group. Conclusion: We can conclude that LS can improve diabetic brain damage with the help of irisin immunoreactivity located in neuronal areas.
