Abstract :
Objective: This study was undertaken to investigate cadmium chloride-induced structural changes and the probable protective effects of co-administration of metallothionein in the testicular tissue.Material Methods: Thirty-six adult Wistar rats were divided in to three groups. Cadmium chloride (3.5mg/kg/day) was subcutaneously injected to the first group (n: 16) whereas second group of rats (n:16) received both cadmium chloride (3.5mg/kg/day) and metallothionein (30μmol/kg/day, ip).The 3rd group (n:4) used as control. On the 1st, 3rd, 5th and 7th day of experimental study, testicular tissue samples were taken under ether anesthesia and fixed with 2.5% glutaraldehyde then prepared blocks for electron microscopic examination.Results: On the electron microscopic examination, particularly after the 3rd day of injection of cadmium chloride, severe tissue damage was evident. The damage gradually increased relative to the time of administration of cadmium chloride. It was determined that cadmium chloride led to collagen increase in the interstitial field, lipid accumulation and apoptosis in the spermatogenic cells increase of mitochondria in the Leydig cells. Similar structural changes were observed in the group that was co-administered metallothionein for protection.Conclusions: As a results, cadmium chloride caused serious structural damage in the testicular tissue. Long term exposure to cadmium chloride may give rise to infertility.
