Effect of Radiation on Rat Uterus Tissue

Abstract :
Objective: The radiological imaging techniques have been widely used for the diagnosis of many diseases, especially in grading and evaluating the response of treatment in malignant conditions. However, the most of these imaging modalities utilize radiation, which is known to result in oxidative damage in tissues. In this study, we aimed to examine the effects of imaging modalities involving the use of radiation on MDA, which is a product of lipid peroxidation, and VEGF immunoreactivity, which is known to be augmented with exposure to radiation. Materials and Methods: Total 20 female Wistar albino rats were divided into 4 groups. Animals in group I comprised the controls. Group II underwent, CT examination at 0 hour with dose of 50 mA and 110 keV, Group III received single dose, 1 mci FDG at 0 hour, followed by a PET/CT examination after 1 hour. Rats in Group IV were administered FDG dose of 1 mci at 0 hour. Animals were decapitated at 24th hour. Uterine tissues were removed using Midsaggital incision and were fixed in 10% formaldehyde for histological examination. After tissues’ routine histological follow-up, they were embedded in paraffin blocks. For MDA and VEGF immunoreactivity, avidin-biotin-peroxidase method was applied to sections. Histoscore based on prevalence and severity of the immune immunoreactivity in the immune staining was created and statistical analysis was performed. Results: In comparison with control rats, there was no observation of significantly differences in VEGF and MDA immunoreactivity in the three other groups. Conclusion: Our results suggest that CT, PET/CT or FDG application has no important effects on VEGF and MDA immunoreactivity in rat uterine tissues
