Abstract :
Pharmacokinetic properties of liposome-encapsulated (LA) and free (FA) ampisilin were compared after intravenous (IV) administrations in sheeps. Six healthy adult sheeps (Akkaraman, female, 15- 18 months aged, weighing 20-24 kg) were used as materials. FA and LA ampicillin were administered IM and IV at single recommended dose (5 mg/kg) to same individual sheeps with 1 weeks interval. Blood samples were obtained at 2.5.,5.,10., 15., 20., 25., 30., 45., 60., 90., 120., 180., 240., 360., 480., 600., 720. minutes after injection. Concentrations of drug in serum were determined by HPLC. Area under curve (AUC= 12.58 μg.saat/ml), elimination half life (t1/2β= 2.28 h), mean residence time (MRT= 1.90 h), and the volume of distribution at steady state (Vd(ss)= 0.78 l/kg) of IV LA was significantly higher than IV FA. Clearance (Cl= 0.41 l/h/kg) of IV LA was lower than IV FA.
