Abstract :
The enzyme responsible for the hydrolysis of glucosinolates is known as myrosinase (E.C. number 3.2.3.2, also known as: β-thioglucosidase, β-thioglucoside glucohydrolase). Some aliphatic glucosinolates, such as sinigrin, glucoraphanin (GRA), and glucoraphenin (GRE) have attracted much attention owing to their myrosinase-mediated breakdown products which have been considered as promising compounds for fighting cancer and the tumor degrading effect of myrosinase enzyme gained importance in recent years. In this study, myrosinase from chicory (Cichorium intybus L.) was purified to homogeneity by ammonium sulfate fractionation, Qsepharose and concanavalin A sepharose affinity chromatography. The purified protein was observed as a single band with a mass of about 35 kDa on SDS-polyacrylamide gel electrophoresis. The enzyme exhibited high activity at pH 8,0 and 70°C. The purified enzyme remained stable at 4 °C for more than 1 year. Using sinigrin as a substrate, the KM ve Vmax values for the purified enzyme were determined to be 0,02 ve 0,074 μmol glucose. min^–1.mg^–1 respectively. The enzyme was strongly activated by ascorbic acid (AA), EDTA, SDS and β-merkaptoethanol. In addition, 10, 1 and 0,1 mM, some effects of ions (Cu^2+, Ni^2+, Ca^2+) on the enzyme activity myrosinaz has been investigated and that they significantly affect the enzyme activity were observed. It was investigated usable of the myrosinaz enzyme obtained from Chicory (Cichorium intybus L.) in treatment of cosmetics In this study, the cream contains pure myrosinaz enzyme is formed and this mixture was applied over 10 volunteer administered that have the skin problems like sun spots for 20 days.The partial reductions in the severity of spots that are available were observed at the end of days specified.
