Abstract :
Ribonucleic acid (RNA) ineterference, is an evolutinaly conserved biyologic process in which double stranded short RNA sequences leads transcriptional suppression of targeted genes. Ever since its discovery it has been widely used in many different areas. In this study, we aim to suppress the active WNT pathway in Acute Lymphoblastic Leukemia (ALL) cell lines by beta-catenin specific siRNA, key molecule of WNT pathway, and eveluate its effects on apoptosis. We were able to suppress beta-catenin expression with a success rate of 60%. Furthermore in MOLT4 cell line, the apoptosis level was 2 fold higher than untreated cells. In conclusion, the active WNT pathway in ALL cell lines can be suppressed by using siRNA and the cells can be lead to apoptosis. siRNA treatments are fast, easy to apply and cost efficient methods for in vitro studies. Nevertheless there is still need for more expriments for its usage in in vivo and clinical applications.
