Abstract :
Behcet s disease is a chronic inflammatory disorder characterized mainly by recurrent oral aphthous ulcerations, genital ulcerations, skin lesions and uveitis. The etiopathogenesis of Behcet s disease is unknown. It has long been postulated that immunological abnormalities, which are possibly induced by microbial pathogens in genetically susceptible individuals, are important in its pathogenesis. According to the results of our previous study for investigating pathogenesis of Behcet s disease by SEREX, autoantibody response to SWAP70 antigen was detected to be associated with vascular involvement among Behcet s disease subgroups. In this study, our aim was to get expression of SWAP70 gene by cloning and determine autoantibody response to this antigen in sera of Behcet s disease patients and healthy control subjects by ELISA. SWAP70 gene was cloned in E.coli M15 (pREP4) cells by pQE-30 vector and autoantibody response to expressed antigen was studied by ELISA in the sera of BD patients and healthy control subjects. Seropositivity in patients and controls was evaluated for identifying SWAP70 antibody response and there was found 10% positivity for SWAP70 in patients but not in controls. Additionally, there was no significant difference in the distribution of SWAP70 antibodies of seropositive and seronegative patients in Behcet s disease subgroups. To further assess and determine the role of SWAP70 antibody in the pathogenesis of Behcet s disease, larger patient groups are required to be studied.
