Abstract :
Chromatin inununoprecipitation (ChiP) sequencing is used in identifmg transcription factor binding sites and in investigating histone modifications and chromatin modifying proteins. In this review, the focus is the use of ChlP sequencing to identify transcription factor binding sites is focused. Identification of genome-wide binding sites of transcription factors are highly important for understanding of transcriptional regnlatory networks underlie biological process causing the development of diseases, evolution and embryonic development. In ChiP sequencing method, DNA fragments to which a transcription factor is binds is obtained from ChlP and are sequenced by using high-throughput sequencing. Although ChiP sequencing method has many advantages over the previous method, there are still challenges that are essential to solve. In this review, these challenges and the strategies that should be applied are discussed.
