Development of Seedling and Germination of Tomato (Lycopersicon esculentum Mill.) Seeds Pre-applied 24-Epibrassinolide Under NaCl Stress Conditions
Abstract :
In this study, the effect of pretreatment with short term exogenous 24-epibrassinolide (24-epiBL) on seed against NaCl stress in M-28 hybrid tomato cultivar (Lycopersicon esculentum Mill.) was investigated. After surface sterilization, seeds was soaking (10-15 seconds) in 24-epiBL solutions of different concentrations (0, 0.5, 1, 1.5 and 2 μM). Seeds were sowed to ½ Murashige-Skoog (MS) medium supplemented with increasing NaCl concentrations (0, 20, 40 60, 80 and 100 mM) and effect of pretraetment with 24-epiBL against NaCl stress was determined evaulating seed germination, seedling development and photosynthetic pigment contents of plants at the end of 17-days. Germination percentage of seeds, seedling fresh weight and dry weight, the average number of leaves per seedling and rooting were negatively affected by NaCl stress. Whereas these parameters were observed to improvement in pretreatment with short term 24-epiBL on seed in certain concentrations of NaCl. Chlorophyll a and carotenoid contents increased until the 80 mM NaCl concentration, whereas they generally decreased at the high salt concentration (100 mM). Pigment contents were increased by pretreatments with 24-epiBL. As a result pretreatment with short term 24-epiBL on seeds is determined to alleviate the deleterious effects of salt stress according to plant growth and pigment content at M-28 hybrid tomato cultivar.
