EFFICIENT SYNTHESIS OF 2,4-DISUBSTITUTED PYRIDO [3,2-d]PYRIMIDINES INVOLVING SNAr AND SUZUKI-MIYAURA CROSS COUPLING REACTIONS
Abstract :
2,4-Disubstituted pyrido[3,2-d]pyrimidines II and III were synthesized starting from the corresponding 2,4-dichloro derivative 1 through SNAr and palladium catalyzed reactions. Various nucleophiles reacted in C-4 position of 2,4-dichloropyrido[3,2-d]pyrimidine 1 by regioselective SNAr. Further, Suzuki-Miyaura cross-coupling reactions on 4-substituted-2-chloro-pyrido[3,2-d]pyrimidines I were performed in high yields.
