Is there any difference between evaluations of fifty cells instead of one hundred cells in studies based on two-dimensional AgNOR measurement?
Abstract :
The NOR, sites of the ribosomal genes, are located on the secondary constrictions of five pairs of acrocentric chromosomes and are stained by silver if they are active. Many studies have been conducted about those regions based on the evaluation of 100 cells. The aim of the study was to detect if there is any difference between the results obtained by evaluation of 50 cells instead of 100 cells. In the current study, six patients with nodular goitre (age range 24-76 years) and ten individuals with cystic nodular goitre (age range 26-69 years), fine needle aspiration biopsy (FNAB) was performed from whose thyroide, were included. Then the biopsy materials were stained for AgNOR evaluation using a specific protocol. At first 50, and then 100 nuclei per individual have been evaluated, and AgNOR count and “Total AgNOR area / Nuclear area” (TNA / NA) values of individual cells were detected for each group. There was no difference between mean AgNOR number obtained from evaluation of first 50 cells (2.256 ± 0.552) and 100 cells (2.243 ± 0.535) (p = 0.600). Moreover, mean „„TNA / NA‟‟ obtained from evaluation of first 50 cells (5.802 ± 1.524) was not statistically different from the mean values of first 100 cells (5.799 ± 1.525) (p = 0.940). It was concluded that, in studies which based on AgNOR count and measurement, evaluation of 50 nuclei instead of 100 nuclei does not effect the sevsitivity or specifity of the method, and does not alter the results of the study, conversily, it provide advantage because of less time requirement.
