Abstract :
This study aimed to determine the intensity of bacterial wilt disease of tomato in Central, Pazar, Turhal, Niksar and Erbaa counties which are main tomato production areas of Tokat. For this purpose surveys were conducted during the 2011 and 2012 growing seasons. For identification of the bacterial isolates, which were isolated from infected plant samples, morphological, physiological and biochemical tests were performed and also serological (ELISA), molecular (PCR) and fatty acid methyl-ester (FAME) analysis were conducted. Based on the survey results, the average disease intensities were 9.64% and 4.98% in 2011 and 2012 respectively. In 2011, the disease prevalences in Central, Niksar, Erbaa, Turhal and Pazar counties were 88.88%, 54.54%, 52.38%, 46.15% and 18.18% respectively. In 2012 the disease prevalence in Erbaa, Central, Turhal, Pazar counties were 61.90%, 52.85%, 47.72%, and 44.44% respectively. Disease was not determine in Niksar. In 2011, 99 bacterial isolates were isolated from infected plant samples. Number of bacterial isolates, obtained from infected plants, were 163 in 2012 survey. 135 bacterial isolates were determined as causal agent of bacterial wilt disease. These bacterial isolates were identified as Clavibacter michiganensis subsp. michiganensis by morphological, physiological and biochemical tests and confirmed by ELISA, PCR and FAME analysis.
