Abstract :
In this study, the effects of cultivar, nutrient media and explant type on calli quality were investigated to provide for production of secondary metabolite in “Kalecik karası”, “Öküzgözü”, “Erciş” and “Cabernet Sauvignon” grape cultivars. Lamina and internodes obtained from the middle part of the shoots were used for an explants source. Calli were planted in the media contained 1.0 μM BAP (6-benzylaminopurine), 0.1 μM 2,4-D (2,4- dicholorophenoxy-acetic acid ), sucrose (2%) and agar (0.8%) and Gamborg B5 solid media. From the ready media, MS was prepared with adding 4.4 g and B5 with adding 3.2 g for per liter water. pH values of both media were fixed as 5.7. Calli were incubated in the darkness with 25°C temperature and sub-cultivated in two times with 21 d interval. As result of this study, it should be stated that type I callus formation indicated for callus quality was changed with media, genotype and explants types. From the two media, B5 was more favorable than MS for the formation of type I callus. The highest and least type I callus formations were observed in “Cabernet Sauvignon” (74.73%) and “Öküzgözü” (23.20%) respectively. As considered two explants, it was observed that lamina was more favorable for the type I callus formation.
