Abstract
von Hippel-Lindau (VHL) gene protein related to tumor suppression, well-defined function is taking part of E3 ubiquitin ligase complex as a substrate recognizer. In normoxia conditions this complex provides rapid proteasomal degradation of , hipoksi indüklenebilir faktör 1-α (HIF-1α) via the ubiquitination by targeting HIF-1α. VHL function aberrations that occurs by mutation or promoter methylation, lead to accumulation of HIF-1α in the cell, so this also activate transcription of a series of hypoxia response genes and so accelerateblood vessel formation that prerequisite for development of a tumor. In this review, we wanted to examine VHL-E3 ubiquitin ligase complex containing tumor suppression and molecular mechanisms that have important roles in the VHL/HIF-1 regulation in oncogenesis.
