The Effects of Different Basal media on Somatic Embryogenesis in Some Alfalfa Cultivars

Abstract
In order to determine the responses of different alfalfa genotypes to plant regeneration through somatic embryogenesis, Prosementi, Gea, Savaş, Ömerbey, OS66, Artemis and Classe alfalfa cultivars were used. Leaflet and cotyledon explants of the cultivars were cultured on different basal media (MS, B5, SH and N6) containing 4 mg/L 2,4-D, 0.5 mg/L kinetin, 2 g/L proline and 30 g/L sucrose solidified with 8 g/L agar. The research was carried out in a randomized plots-factorial experimental design with 3 replications and 10 explants per replication. As a result of the research, callus weight per petri dish, the number of somatic embryos and plantlets per petri dish varied respectively between 0.253-6.700 g, 0-19.667 and 0-6.667. The maximum number of somatic embryos and plantlets per petri dish was obtained from Ömerbey alfalfa cultivar cultured on SH basal medium in the leaflet explant; whereas in the cotyledon explant, the highest number of embryos per petri dish was obtained from N6 medium in OS 66 cultivar and no plantlet regeneration was achieved from cotyledon explants.
