Abstract
This paper experimented with the λ-DNA molecules which were successfully stretched on the hydrophobic surface of PMMA by the molecular combing method in a wide concentration range from 7mM to 20mM of tris buffer. The buffer concentration has a remarkable influence on the properties of the stretched DNA such as the length and uniformity. The results demonstrated the most applicable concentration of tris buffer for λ-DNA stretching is 10mM.
