Abstract :
Our research group has purified an antioxidant selenium-containing protein (Se-GL-P) from Se-enriched Ganoderma Lucidum (Se-GL), its characteristics has been determined partly. In this study, the characteristics of Se-GL-P were identified further, in which the contents of selenocysteine (Se-Cys) and selenomethionine (Se-Met), isoelectric point (pi), and peptide mass fingerprint (PMF) were involved. Moreover, in vitro, the effects on multiplication of T and B lymphocyte cell and killing activity of NK cell were determined by method of MTS. The effect on secretion of TNF-alpha was determined by method of ELISA. Results showed that Se-GL-P was a kind of new selenium-containing protein proved by method of peptide mass fingerprint. 31.275 % of Selenium in total selenium amino acids was in form of Se-Cys, and 68.725% of that was in form of Se- Met. All groups with different doses of Se-GL-P could promote the multiplication of T and B lymphocyte cell of mouse as well as promoting the killing activity of NK cell. However, Se-GL-P had no better effect on TNF-alpha secretion by macrophages in mouse. At the same concentration, the immune regulation activity of Se-GL- P was higher than crude protein from Se-GL (SeGLPr). The immune regulation activity of the former was almost 1.5 times of the latter. Consequently, the results indicated that the Se-GL-P could be studied further, as a kind of bioactivity component with high immune regulation activity.
