Abstract :
Streptococcus pneumoniae is the major causative organisms of meningitis, bacteremia, and pneumonia. The drug resistance of S.pneumoniae to antibiotics showed ascending tendency. The present study cloned the full-length HMG-CoA reductase (HMGR) gene of S.pneumoniae and expressed in E.coli BL21 (DE3). The His-tag containing recombinant protein was purified by Ni-NTA affinity chromatography. Statins represent a kind of excellent competing inhibitor targeted for the class I HMG-CoA reductase, but were relatively poor to the class II enzyme of important bacterial pathogens. In order to develop new inhibitors with more specific and stronger function targeted for the class II HMG-CoA reductase, the 3D model of S.pneumoniae HMG-CoAR was built based on structure template of HMGR of Pseudomonas mevalonii by homology modeling using composer module of SYBYL7.0 program.
