Abstract :
HPLC was used to separate chiral compounds with chiral column. Another method was used in HPLC with chiral mobile phase additives to separate chiral compounds. Using chiral mobile phase additives is a good way to separate the chiral compounds. The structure of the derivative takes an important role in separation. We derivatized 2-heptamine with benzene sulfonyl chloride and dansyl chloride, Using cyclodextrin derivatives as chiral mobile phase additives, 2-heptamine derivatives was separated by HPLC. 2-heptamine derivatives have a good absorbstance under 254 nm. We separated these derivative with chiral mobile phase additives, the additives were beta-CD, 2-O-(2,3-dihydroxypropyl)-beta-CD, 2-O-(hydroxypropyl)-beta-CD and so on. The additives are all highly hydro-soluble and good for separation. We used phosphoric buffer solution to supply proper pH range and ion intensity. A good separation result was obtained using 2-O-(hydroxypropyl)-beta-CD. It could be used in separation of molecule like aliphatic amine. Benzene sulfonyl chloride and dansyl chloride could be used to derivatize 2-heptamine for its chiral separation. And we got some conclusion about how to separate alkyl amine, and how to select the chiral mobile phase additives. We got a mechanism that the role chiral mobile phase additives played during the separation.
