Abstract :
The concept of biosensor based on imaging ellipsometry was proposed in 1995. Imaging ellipsometry is used for the acquisition of the bio-molecule interaction information between biomolecule analytes and ligands on a solid substrate. Due to the phase sensitive property of imaging ellipsometry and a coupling with a multifunction microfluidic array reactor, the biosensor approach has been realized with some properties of the fast sampling, label free, high throughput, and real-time. Besides the imaging ellipsometry, a series of related techniques have been developed for the biosensor, which mainly includes the surface pattern fabrication of a protein microarray, the various ligand oriented immobilization, the samples delivery, the biomolecule affinity presentation, the specific interaction, the prevention of unspecific binding, as well as the realization of the detection sensitivity of 0.1 ng/mL for immunoglobulin, the sample consumption about 10 L, the sampling and calibration for quantification, etc. It has been developed into an automatic analysis technique for protein detection. A 48 independent channel throughput protein analysis. Its principle, methodology, related techniques, especially how to realize the biosensor system and its biomedical applications is presented in this report.
