  • Abstract
    Poliomyelitis though well controlled in developed countries, is still a major problem in developing countries. Causal agent, a virus belonging to family Picornaviridae, occurs in 3 serotypes. In the virus, neutralization antigenic sites (N-Ags) are located on capsid proteins. Antibody secretion helps in recovery from specific viral diseases, which are present in the serum or extracellular body fluids or may be introduced artificially. An effort has been made to deduce 3-dimensional structure of VP1 protein of type 3 poliovirus and a complete amino acid sequence of Sabin vaccine derivative P3/LEON/12a1b, which will help to prepare a basis for improvement in designing and enhancing the efficacy of the structure based vaccine. The amino acid sequence of VP1 protein was obtained from NCBI databank. Its homology was studied and template selected from PDB using BLAST. Sequence alignment was done using ClustalX and 3-dimensional modeling was carried out using Modeller8v0. The structure was visualized in Rasmol and refined in VMD 1.8.3. Finally Ramchandran plot formed the basis of the protocol, aiming to deduce 3-dimensional structure of one of the key target proteins of Poliomyelitis virus to precisely understand the vaccine
