Abstract :
RNA interference has unveiled the hidden sound of silence in many organisms today. To follow suit, an intron-containing hairpin RNA (ihpRNA) plasmid vector, Psilent -1, with inverted repeats of size ~800bp was constructed. Rapid amplification of complementary DNA ends (RACE) revealed the 3´-full length of ERG 2 gene coding region as 989bp. This encodes 321 amino acids of molecular weight 33.89kDa and a theoretical isoelectric point of 5.05. This silencing construct is the underlying bedrock upon which work in the near future concerning ergosterol biosynthetic pathway and expression in C. cupreum at molecular level will richly overlay.
