Abstract :
The effect of the different substrate concentration, enzymatic compositions, varied chemical activators on the cellulase hydrolysis process was studied. The results showed that(1) the effect of the enzymatic hydrolysis was vital when the substrate was more than 5% (2) the enzymatic composition had much more effect ont the yield of cellulase hydrolysis(3) Mg2+, Co2+ had obvious effect in the low concentration (<;1mM/L) with increasing 30 g/L compared to blank sample(4) the surfactants PEG 2000 and Tween 80 also had notable effect on the enzymatic hydrolysis with increasing 23 g/L. The reduce sugar was obtained up to 115g/L with glucose concentration 88g/L when the enzyme was loaded according to U/g cellulose FPA 45: β-glucodase 25: xylase 800 with 20% substrate concentration, 120-150rpm, pH4.8, 50°C for 144h. The converting yield was up to 85%. The study on the the characterization of cellulase hydrolysis in the high-solid substrate had an important significance for the bioprocess of ligninocellulose.
