Abstract :
To establish the RP-HPLC method for simultaneous determination of two effective constituents in different parts of Juglans mandshurica from various sources. Two effective constituents were simultaneously determined by the RP-HPLC with WATERS XBridge C18 column (4.6 mm×250 mm, 5 μm ) by gradient elution in methanol and water as the mobile phase. The flow rate was 1.0 mL· min-1; the detection wavelength was 255 nm with column temperature at 25 °C. Two effective constituents were separated clearly and respectively in 35 min, the linear ranges of juglone and pinostrobin were 0.00066~0.033 μg (r=0.9997) and 0.054~2.7000 μg (r=0.9996), respectively; the average recoveries were 97.15% and 97.82%, respectively. RSD% were 1.41% and 1.96%.The method is simple, accurate and can be used for quality control and evaluation of Juglans mandshurica.
