Abstract :
A kind of polymeric prodrug of ibuprofen with glucose pendant was prepared through chemo-enzymatic method. The obtained polymeric prodrug was characterized by FT-IR, NMR and GPC. The molecular weight of the polymeric prodrug reached 2.126 × 104. Furthermore, the in vitro release behaviors of ibuprofen (raw drug) and the polymeric prodrug were investigated in pH 7.4 buffer solution at 37°C. It was suggested that low-molecular-weight ibuprofen was fast released and the cumulative release amount was almost 100% after 3 h, while only about 4 % of parent drug was released from polymeric prodrug in the same time. The continuous release investigation showed that the cumulative parent drug amount released from the polymeric prodrug reached 32% after 10 days. Compared to the raw drug, the polymeric prodrug of ibuprofen with glucose pendant had a better effect of prolonged release, and the release of ibuprofen from polymeric prodrug exhibited first order kinetic equation.
