Abstract :
To detect and identify of Enterohemorrhagic Escherichia coli O157:H7, the gene rfbE of O157:H7 was detected as the aim gene and the Agilent 2100 bioanalyzer was used as an analysis tool, which was compared with the conventional agarose gel electrophoresis. Finally, it was known that the rfbE gene was successful in detection of O157:H7. And the bioanalyzer was as 10 times sensitive as the agrouse gel electrophoresis; at the same time it could give the accurate size, whose error rate was less than 5%. The conc and molarity of the different Cell concentration could be also provided. The bioanalyzer has the potential to replace the gel electrophoresis, but improvements in the chip separation are needed. Overall, the sensitivity and reproducibility of the agilent 2100 bioanalyzer make it a valuable tool for the analysis of special DNA fragments in clinical samples, which could aid in the development of diagnostic parameters with respect to enterohemorrhagic Escherichia coli O157:H7 detection.
