Abstract
This paper presents new algorithm of plant nuclei segmentation for the purpose of DNA contents measurements. This algorithm is based on preliminary discrimination of nuclei objects in RGB colour space and then on local applying of some global thresholding methods in a rectangular ROI area. Segmented nuclei objects are zoomed more than in typical applications to improve quantitative analysis
