Purpose: To optimize expression and purification conditions of 3α Hydroxysteroid Dehydrogenase (3α-HSD) to prepare highly pure and active interest protein; Methods: To find optimal expression conditions according to the optimization of expression conditions; to find optimal purification conditions according to comparison between the effects of different purification conditions; to obtain optimal preservation of enzyme according to comparison between the effects of different conditions on enzymic activity. Results: The induction expression of protein reaches the peak after 6 hours; the optimization of purification conditions shows that it is the best purification method on economic effects to combine ammonium sulfate precipitation and ion exchange; 3α-HSD is stable under the conditions of less than 25 degree and pH9. Conclusions : To find optimal preparation conditions for 3α-HSD and obtain optimized preservation conditions for enzyme according to optimization of conditions of expression and purification.
