RAPD markers were used to analyze genetic stability of regenerated plant from different ways of Populus hopeiensis in this study. The results indicated that, using the young leaves of vitro plantlets as explants, the RAPD bands were same between the control and regenerated plant which was from the direct regenerating adventitious buds. The young leaves of vitro plantlets were as explants to induce the callus, and there was a polymorphic band respectively in the primer S268 and S104 on 2 regenerated plants which were from the way of organogenesis. The mutational rate of RAPD sites and plants were 2/288 and 2/20 respectively. It indicated that there was somaclonal variation during training by this way, and the genetic stability was lower than by the former way.
