Trichlorfon (TCF) is a widely used broad-spectrum agricultural organophosphorus (OP) pesticide. Few studies have evaluated the effects of TCF on reproductive toxicity after low level exposure, especially long-term exposure. In this study we assessed the direct effects of TCF on estrous cycle in female mice and developmental outcomes in near-term fetuses after consecutive 30 days of maternal exposure to 2, 10 and 50 mg/kg body weight/day TCF via drinking water. Both male and female fetuses in the 50 mg/kg/day TCF treated group had significantly reduced body weight but not in 2 mg/kg/day and 10 mg/kg/day. There was no difference in estrous cycle between the control and TCF-treated groups. No increased incidence of fetal external malformations was observed in the TCF-treated groups. The lack of effects of TCF of 2 mg/kg/day and 10 mg/kg/day on any in vivo reproductive and developmental endpoints above suggested that for TCF, a hazard of reproductive toxicity below 10 mg/kg/d may not be expected. However, a significant decrease in fetal body weight was observed in the 50 mg/kg/day TCF treated group, suggesting that potential adverse health effects of OP pesticides exposure should continue to be carefully studied in the future.
