Abstract :
Ribosomal protein S3A (RPS3A) is a component of the 40s small ribosomal subunit, which plays an important role in ribosome. To explore the structure characteristic of RPS3A gene of giant panda (Ailuropoda Melanoleuca), the cDNA and the genomic sequence of RPS 3A were cloned successfully from the giant panda by RT-PCR and Touch-down technology respectively, followed by sequencing and analysing. The length of cDNA fragment cloned was 839bp, containing an open reading frame of 795bp. The deduced protein was composed of 294 amino acids with an estimated molecular weight of 32.89KD and PI of 4.51. The genomic sequence is 3926 bp in length, possesing six exons and five introns. Alignment analysis indicated that the homologies for nucleotide sequence of giant panda RPS 3A with that of Homo sapiens, Bos Taurus, Mus musculus, Rattus norvegicus and Sus scrofa are 93.58%, 93.96%, 88.68%, 89.43% and 93.71%, while for amino acids the homologies are 100.00%, 100.00%, 99.24%, 99.24% and 98.11% respectively. Topology prediction showed that there are six different patterns of functional sites in the RPS3A protein of giant panda. The results will enrich and supplement the mammals RPS3A gene database.
