Abstract :
Under flow conditions, neutrophil attachment to the endothelial lining of the blood vessel is mediated by various combinations of L-, P-, and E-selectin. When ordered in close succession, the formation of transient selectin bonds by neutrophils results in rolling on the endothelium. Recent reports and biophysical modeling studies have suggested that selectin mediated rolling requires a relatively low reactive bond compliance compared to antigen-monoclonal antibody interactions. Based on in vivo and in vitro evidence that L-selectin mediated interactions. may be on the order of standard video capture rates, we directly measured selectin tethers at high temporal resolution comparing their kinetics and reactive compliance at similar site densities
