Relationship between protective effect of hesperidin extraction on exercise fatigue and antioxidation

Abstract :
To explore the antioxidative effect of hesperidin extracted from the peel of Gannan kumquat on exercise fatigue. Methods: Hesperidin was extracted from the peel of kumquat by ultrasound and ethanol precipitation and was detected qualitatively by the high performance liquid chromatography. Animal model of exercise fatigue was established by the 10-day increasing progressively swimming training. Effect of hesperidin on serum glutathione peroxidase, superoxide dismutase, catalase, myeloperoxidase and maleic dialdehyde of exercise-fatigue rats was tested by the spectrophotometric method. Results: The rats of exercise fatigue had some phenomena, such as depilation, tired appearance, low appetite, degressive locomotory capacity and slow-moving growth of body weight. Exercise fatigue caused the rats to decrease the enzyme activity of serum GSH-PX (P<;0.001) and to increase the enzyme activity of serum MPO (P<;0.05); however, exercise fatigue had no significant difference on the enzyme activity of serum SOD and CAT, and concentration of MDA (P>;0.05). The rats of intragastric administration by extractive hesperidin had fine appetite, glossy hair, persistent growth of body weight. Hesperidin had no positive and negative function on the enzyme activity of serum SOD and CAT of exercise fatigue rats (F=0.4589, P>;0.05; F=0.8556, P>;0.05). Low, middle and high dose of hesperidin had no influence on the enzyme activity of serum MPO (P>;0.05); nevertheless, low, middle and high dose of hesperidin increased the enzyme activity of serum GSH-PX (P<; 0.001) although it couldn´t recover the normal level of enzyme activity of serum GSH-PX. At the same time, hesperidin decreased the concentration of serum MDA in exercise fatigue rats (P<;0.001). Conclusion: Hesperidin had inhibited the exercise fatigue through increasing the enzyme activity of serum GSH-PX, decreasing the concentration of serum MDA.
