چكيده فارسي :
زمينه و هدف: سرطان ريه شايع ترين بدخيمي و اولين عامل مرگ و مير ناشي از سرطان در ميان زنان و مردان جهان است، به طوري كه بقاي 5 ساله بيماران حدود 15% مي باشد. با توجه به نقش تهاجمي CXCL12 و چالش هاي موجود در مطالعات مختلف در بيان و عدم بيان CXCL12 و وجود متاستاز در مراحل اوليه ي آدنوكارسينوماي ريه، رده سلولي A549 انتخاب شد تا ميزان بيان ژن CXCL12 در آن مورد بررسي قرار گيرد.
روش كار: در اين مطالعه رده سلولي A549 در محيط كشت DMEM حاوي 10% سرم گاوي و 1% پني سيلين-استرپتومايسين كشت داده شد. از رده ي سلولي مذكور RNA استخراج گرديد و پس از ارزيابي كمي و كيفي آن،cDNA سنتز شد. سپس ميزان بيان ژن CXCL12 با روش Real Time PCR مورد ارزيابي قرار گرفت.
يافته ها: بيان اندك CXCL12 در رده سلولي آدنوكارسينوماي ريه A549 نشان دهنده ي آن است كه CXCL12 مي تواند به عنوان يك ماركر بالقوه براي اهداف پيشگويي كننده و در نهايت درماني در نظر گرفته شود.
نتيجه گيري: CXCL12 در رده سلولي آدنوكارسينوماي ريه A549 بيان كمي دارد.

Inroduction & Objective: Lung cancer is the most common malignancy and the leading cause of cancer related mortality among women and men worldwide, in such a way that the 5-year survival of patients is about 15%. According to the invasive role of CXCL12, challenges in various studies about expression and non-expression of CXCL12 and the presence of metastasis in early stages of lung adenocarcinoma, the A549 cell line is selected in order to investigate CXCL12 gene expression in it.Material and Methods:In this study A549 cell line was cultured in a DMEM medium contain 10% fetal serum bovine and 1% penicillin- streptomycin. RNA extraction was performed from the mentioned cell line and after qualitative and quantitative evaluation of extracted total RNA, cDNA was synthesized. Then CXCL12 expression was measured using Real time PCR.
Results: CXCL12 low expression in lung adenocarcinoma A549 Cell line shows that it can be considered as a potential markerfor predictive and therapeutic purposes.Conclusion:CXCLL12 has a low expression in A549 cell line.
