Introduction Isolation and Identification Methods of Eight Form-Species Trichoderma

شبه گونه هاي Trichoderma از مهم ترين قارچ هاي خاك زي متعارض قارچ هاي بيمارگر گياهان مي باشند، كه در القاي مقاومت در گياهان زراعي نيز نقش دارند، بنابراين جداسازي و شناسايي آن ها مي تواند كمك زيادي به استفاده از آن ها در مبارزه زيستي با بيمارگرها نمايد. اين قارچ ها را مي توان با روش رقيق سازي سوسپانسيون خاك و كشت روي محيط هاي داوه، پپتون رزبنگال و يا سيب زميني/دكستروز/آگار تعديل شده، جداسازي كرد. شناسايي آن ها نيز بر اساس ويژگي هاي پرگنه ها، خصوصيات ريخت شناسي كنيديوم برها، فياليدها و كنيديوم ها صورت مي-گيرد. هشتادو پنج جدايه ي Trichoderma از 25 نمونه ي خاك، 8 منطقه ي چغندركاري استان فارس به اين روش جدا و خالص سازي شدند. با مطالعه صفات آن ها، 8 شبه گونه ي Trichoderma ،به اسامي T. asperellum، T.atroviride، T. brevicompactum، T. harzianum، T. longibrachiatum، T. spirale،T. tomentosum و T. virens ، شناسايي شدند. روش هاي جداسازي، شناسايي و خصوصيات ريخت شناسي اين قارچ ها شرح داده شده است.

Trichoderma form-species are among of the important antagonists of plant pathogenic fungi and also induce resistance in field crops. Therefore their isolation and identification could be helpful for applying them in biological control. They isolate by pour soil diluted suspension on Dawet, Peptone – Rose Bengal, or modified Potato- Dextrose-Agar mediums. Species can identify based on colony characteristics, growth characteristics and morphological characteristics of conidiophores, phialids, and conidia. Eighty-five isolates of Trichoderma from 25 soil samples, from 8 regions of the Fars province sugar beet fields, isolated and purified with this method. With studied their characteristics eight form-species of Trichoderma identified with names: T. asperellum, T. atroviride, T. brevicompactum, T. harzianum, T. longibrachiatum, T. spirale, T. tomentosum and T. virens . Methods of isolation, identification and morphological characteristics of these fungi described.
