بررسي اثرات بيهوش كنندگي بيكربنات سديم، 2-فنوكسي اتانول، عصاره گل ميخك و عصاره آويشن بر شاخصهاي خون شناسي و ميزان هورمون كورتيزول در ماهي كپور معمولي (Cyprinus carpio)

اثرات بيهوش‌كنندگي دو تركيب شيميايي بيكربنات سديم و 2-‌فنوكسي اتانول و دو تركيب طبيعي عصاره گل ميخك و عصاره آويشن بر تعداد گلبولهاي قرمز و سفيد و مقدار هموگلوبين، هماتوكريت و هورمون كورتيزول خون مورد آزمايش قرار گرفت. در اين خصوص شاخصهاي خونشناسي در ماهيان بيهوش شده با 30 ميليگرم در ليتر عصاره گل ميخك، 100 ميليگرم در ليتر عصاره آويشن، 30 ميليگرم در ليتر بيكربنات سديم و 3/0 ميليليتر در ليتر2 - ‌فنوكسي اتانول و يك گروه شاهد بدون ماده بيهوشي در دو زمان 10 دقيقه و 24 ساعت پس از بيهوشي بررسي گرديد. نتايج بدست آمده كاهش در تعداد گلبول قرمز و گلبول سفيد و مقدار هموگلوبين و هورمون كورتيزول و افزايش در مقدار هماتوكريت در تيمارهاي آويشن و عصاره ميخك در 10 دقيقه پس از بيهوشي را نشان داد. همچنين افزايش معني‌دار در تعداد گلبول قرمز، گلبول سفيد و مقدار هماتوكريت و هورمون كورتيزول خون در تيمار 10 دقيقه و 24 ساعت پس از بيهوشي با بيكربنات سديم نسبت به شاهد مشاهده شد (05/0P<). اما مقدار هموگلوبين خون در تيمار 24 ساعت پس از بيهوشي با بيكربنات سديم، عصاره گل ميخك، عصاره آويشن و 2-‌فنوكسي اتانول كاهش معني‌دار نشان داد (05/0P<). در مورد تيمار 2- فنوكسي اتانول اگرچه تعداد گلبولهاي قرمز و سفيد و مقدار هماتوكريت در 10 دقيقه افزايش يافت اما در زمان 24 ساعت روند كاهشي مشاهده شد و تفاوت با گروه شاهد معنيدار نبود. بنابر اين كمترين ميزان استرسزايي مربوط به عصاره آويشن و عصاره گل ميخك بود. و در تيمار 2- فنوكسي اتانول نيز اثرات منفي غير قابل بازگشت مشاهده نشد.

In order to introduce an anaesthetic with lower stressful effects on common carp, the effect of anaesthesia with two chemical compositions, sodium bicarbonate and 2-phenoxyethanol and two natural materials, clove extract and thyme extract on the number of red and white blood cells (RBC, WBC) and the amount of hematocrit, hemoglobin (HCT, HB) and Cortisol hormone were studied. In this regard, the hematological indices were evaluated in fish anesthetized with 30 mg/l clove oil, 100 ppm thyme extract, 30 mg/l of sodium bicarbonate and 0.3 ml/l 2-phenoxyethanol and a control group with no anesthesia, at the time 10 minutes and 24 hours after anesthesia. The results showed a significant decrease in the number of RBC, WBC and the amount of HB and cortisol and an increase in the amount of HCT in clove extract and thyme extract, 10 min after anesthesia. Also, a significant increase was observed in the number of RBC and WBC and the amounts of hematocrit and cortisol in sodium bicarbonate treatment, compare to the control group, 10 min and 24 hours after anesthesia (P<0.05). But a significant decrease was found in the amount of hemoglobin in all treatments, 24 hours after anesthesia (P<0.05). About 2-phenoxyethanol, although the numbers of RBC and WBC and the amount of HCT increased 10 min after anesthesia but, it showed decreasing trend with no difference compare to the control group, 24 hours after anesthesia. So, the lower stressful effects were belonged to clove extract and thyme extract and the negative irreversible effects were not observed in 2- phenoxyethanol treatment.
