چكيده فارسي :
ﺳﺎﺑﻘﻪ و ﻫﺪف: ﻣﺴﯿﺮ Tweak /Fn14 ﭘﺎﺳﺦﻫﺎي ﻓﯿﺰﯾﻮﻟﻮژﯾﮑﯽ ﻣﺨﺘﻠﻒ ﺷﺎﻣﻞ ﺑﻘﺎ ﺳﻠﻮل، ﺗﮑﺜﯿﺮ، ﺗﻤﺎﯾﺰ، رگزاﯾﯽ، ﻣﻬﺎﺟﺮت و آﭘﻮﭘﺘﻮز را ﺗﻨﻈﯿﻢ ﻣﯽﮐﻨﺪ. ﻫﺪف ﭘﮋوﻫﺶ ﺣﺎﺿﺮ ﺑﺮرﺳﯽ اﺛﺮ ﻟﯿﮕﺎﺳﯿﻮن ﻋﺼﺐ ﻧﺨﺎﻋﯽ ﺑﺮ ﺑﯿﺎن ژنﻫﺎي Tweak و Fn14 ﻋﻀﻠﻪ EDL ﻣﻮشﻫﺎي ﻧﺮ وﯾﺴﺘﺎر ﭘﺲ از ﺗﻤﺮﯾﻦ HIT ﺑﻮد.
ﻣﻮاد و روشﻫﺎ: ﺑﺪﯾﻦ ﻣﻨﻈﻮر، 24 ﻣﻮش ﻧﺮ وﯾﺴﺘﺎر در 2 ﮔﺮوه 12 ﺗﺎﯾﯽ ﮐﻨﺘﺮل (C) و ﺗﻤﺮﯾﻦ (HIT) ﻗﺮار ﮔﺮﻓﺘﻨﺪ. ﮔﺮوهﺗﻤﺮﯾﻦ ﺑﻌﺪ از ﯾﮏ ﻫﻔﺘﻪ آﺷﻨﺎﺳﺎزي ﺑﺮﻧﺎﻣﻪ 4 ﻫﻔﺘﻪ HIT را اﻧﺠﺎم دادﻧﺪ. ﺳﭙﺲ ﺑﻪ ﻃﻮر ﺗﺼﺎدﻓﯽ ﺳﺎده ﺑﻪ دو ﮔﺮوه 6 ﺗﺎﯾﯽ ﺗﻤﺮﯾﻦ (HIT) و ﮔﺮوه ﻟﯿﮕﺎﺳﯿﻮن ﻋﺼﺐ ﻧﺨﺎﻋﯽ (HIT-SNL) ﻗﺮار ﮔﺮﻓﺘﻨﺪ. ﻫﻤﺰﻣﺎن ﮔﺮوه ﮐﻨﺘﺮل ﻧﯿﺰ ﺑﻪ دو ﮔﺮوه 6 ﺗﺎﯾﯽ ﮐﻨﺘﺮل (C) و ﻟﯿﮕﺎﺳﯿﻮن ﻋﺼﺐ ﻧﺨﺎﻋﯽ (C-SNL) ﺗﻘﺴﯿﻢ ﺷﺪﻧﺪ. 4 ﻫﻔﺘﻪ ﭘﺲ از SNL ﻣﻮشﻫﺎ ﻗﺮﺑﺎﻧﯽ و ﻋﻀﻠﻪ EDL و ﻧﻌﻠﯽ اﺳﺘﺨﺮاج و ﺑﯿﺎن ژنﻫﺎ ﺑﻪ روش ﮐﻤﯽﺳﺎزي دادهﻫﺎ Real-time PCR اﻧﺪازهﮔﯿﺮي ﺷﺪ و ﺑﺎ اﺳﺘﻔﺎده از آزﻣﻮن ﺗﺤﻠﯿﻞ وارﯾﺎﻧﺲ دو راﻫﻪ دادهﻫﺎ ﻣﻮرد ﺗﺠﺰﯾﻪ و ﺗﺤﻠﯿﻞ ﻗﺮار ﮔﺮﻓﺖ. ﯾﺎﻓﺘﻪﻫﺎ: ﻧﺘﺎﯾﺞ ﻧﺸﺎن داد ﮐﺎﻫﺶ ﻓﻌﺎﻟﯿﺖ ﺑﺪﻧﯽ ﺑﻪ ﺷﮑﻞ SNL ﺳﺒﺐ اﻓﺰاﯾﺶ ﻣﻌﻨﯽداري ﺑﯿﺎن ژنﻫﺎي Tweak وFn14 ﻣﯽﺷﻮد (ﺑﻪ ﺗﺮﺗﯿﺐ 0/0001=P و 0/002=P). ﻫﻤﭽﻨﯿﻦ اﻧﺠﺎم ﺗﻤﺮﯾﻦ HIT ﻗﺒﻞ از SNL در ﮔﺮوه ﺗﻤﺮﯾﻦ- SNL ﺑﻪﻃﻮر ﻗﺎﺑﻞ ﻣﻼﺣﻈﻪاي ﺑﯿﺎن ژنﻫﺎي Tweak و Fn14 را ﻧﺴﺒﺖ ﺑﻪ ﮔﺮوه ﮐﻨﺘﺮل-SNL ﮐﺎﻫﺶ ﻣﯽدﻫﺪ (ﺑﻪ ﺗﺮﺗﯿﺐ 0/0001=P و 0/001=P). ﻋﻼوه ﺑﺮ اﯾﻦ، ﯾﺎﻓﺘﻪﻫﺎ ﻧﺸﺎن ﻣﯽدﻫﺪ SNL اﺛﺮ ﻣﻌﻨﯽداري ﺑﺮ ﻧﺴﺒﺖ وزن ﻋﻀﻠﻪ ﻧﻌﻠﯽ ﺑﻪ ﻃﻮل درﺷﺖﻧﯽ دارد( 0/01=P). از ﻃﺮف دﯾﮕﺮ اﻧﺠﺎم ﺗﻤﺮﯾﻨﺎت HIT ﻗﺒﻞ از SNL ﺳﺒﺐ اﻓﺰاﯾﺶ ﻣﻌﻨﯽدار آن ﺷﺪ (0/03=P).
ﻧﺘﯿﺠﻪﮔﯿﺮي: ﮐﺎﻫﺶ ﻓﻌﺎﻟﯿﺖ ﺑﺪﻧﯽ ﺑﻪ ﺷﮑﻞ ﻟﯿﮕﺎﺳﯿﻮن ﺑﺎ اﻓﺰاﯾﺶ ﺑﯿﺎن ژنﻫﺎي Tweakو Fn14 ﻫﻤﺮاه ﺑﻮد.از ﻃﺮف دﯾﮕﺮ ﺗﻤﺮﯾﻦ ﺗﻨﺎوﺑﯽ ﺷﺪﯾﺪ در ﮐﺎﻫﺶ ژنﻫﺎي آﺗﺮوﻓﯿﮏ ﻣﻮﺛﺮ ﺑﻮده اﺳﺖ. ﺑﺎ ﺗﻮﺟﻪ ﺑﻪ اﯾﻦ ﯾﺎﻓﺘﻪﻫﺎ، اﻧﺠﺎم ﺗﻤﺮﯾﻨﺎت HIT ﻗﺒﻞ از اﺑﺘﻼ ﺑﻪ ﺑﯽ ﻓﻌﺎﻟﯿﺘﯽ ﺑﺘﻮاﻧﺪ روﻧﺪ آﺗﺮوﻓﯽ ﻋﻀﻼﻧﯽ را ﮐﺎﻫﺶ دﻫﺪ.
چكيده لاتين :
Background and Purpose: The Tweak/Fn14 pathway regulated various physiological responses including cell survival, proliferation, differentiation, angiogenesis, migration and apoptosis. The purpose of the present study was to investigate the effect of SNL on the expression of Tweak and Fn14 genes in the EDL muscle of Wistar rats after HIT training.
Materials and methods: For this purpose, 24 male Wistar rats were divided into 2 groups of control (C) (n=12) and training (HIT) (n=12). After one-week familiarization, training group participate in four HIT training. Then, they were randomly assigned to HIT (n=6) and HIT-SNL (n=6) groups. At the same time, the control group was divided into two groups: control (C) (n=6) and SNL (C-SNL) (n=6). 4 weeks after the SNL, wistar rats were sacrificed then EDL and soleuse muscle exetracted.Then gene expressions of Tweak and Fn14 measured with Real time PCR technique. Data were analyzed using 2-way ANOVA.
Results: The results showed that inactivity via SNL has a significant increased of the expression of Tweak and Fn14 genes (P=0.000and P=0.002, respectively). Also, performed before SNL reduced the expression of Tweak and Fn14 genes in SNL-T compared with SNL-C (P=0.0001 and P=0.001 respectively). In addition, the findings indicated that SNL had a significant effect on the weight of soleuse muscle mass/ tibia length (P=0.01). On the other hand, HIT training before SNL significantly increased ratio weight soleuse muscle mass/ tibia length (P=0.03).
Conclusion: The decrease in activity by SNL is associated with increased in expression of Tweak and Fn14 genes. On the other hand, HIIT training have been effective in reducing atrophic genes. According to these findings, performing HIT training before inactivity can reduced the muscular atrophy process.
