بررسي فعاليت ضد ميكروبي عصاره گياه Scabiosa Olivieri و تاثير آن بر بيان TNF-α و IL-1 در سلولهاي خوني محيطي انسان

Antimicrobial Activity of Scabiosa Olivieri Extract and Its Effect on TNF-α and IL-1 Expression in Human Peripheral Blood Cells (PBMCs

بيماريهاي عفوني ايجاد شده توسط باكتريها و قارچها ميلياردها انسان را در سراسر جهان تحت تاثير خود قرار داده است. در طول تاريخ بشريت، بيماريهاي عفوني عامل اصلي مرگ و مير و معلوليت بوده است. امروزه بيماريهاي عفوني عامل يك سوم تمام مرگها در دنيا مي باشند. هدف كلي اين مطالعه بررسي گياه دارويي Scabiosa Olivieri از نظر خاصيت ضد ميكروبي بر روي باكتري هاي گرم مثبت، گرم منفي و قارچ و نيز مطالعه خاصيت ضدالتهابي آن با بررسي فاكتورهاي IL-1 و TNF-α انساني مي باشد كه در نهايت منجر به معرفي يك عامل ضدميكروبي و ضدالتهابي جديد با منشا طبيعي و ارزان شود. مواد و روش ها: ابتدا عصاره گياه به روش ماسراسيون تهيه گرديد و سپس به منظور بررسي خواص ضدميكروبي، رقت هاي متفاوتي از عصاره اين گياه بر روي سه ميكروارگانيسم اشرشيا كلاي، استافيلوكوكوس اورئوس و كانديدا آلبيكانس انجام شد همچنين تاثير اين عصاره بر روي سلول هاي تك هسته اي خون محيطي و اندازگيري فاكتورهاي التهابي IL-1 و TNF-α بوسيله كيت الايزا مورد بررسي قرار گرفت. نتايج عصاره گياه Scabiosa Olivieri به طور معني داري خاصيت ضدالتهابي از خود نشان داد و همچنين خاصيت ضد ميكروبي فعالي بر روي باكتري اشرشيا كلاي و ضدميكروبي متوسطي بر استافيلوكوكوس اورئوس دارد. اما خاصيت ضدقارچي مشاهده نشد. نتيجه گيري: عصاره اين گياه پتانسيل استفاده بعنوان يك عامل ضدالتهابي و نيز ضدميكروبي را داراست كه البته بررسي اثرات استفاده آن در محيط in vivo و نيز آناليز شيميايي تركيبات تشكيل دهنده آن نيازمند انجام تحقيق هاي بيشتري مي باشد.

Infectious diseases caused by bacteria and fungi have affected billions of people worldwide. Throughout human history, infectious diseases have been the leading cause of death and disability. Infectious diseases today account for one-third of all deaths in the world .The general objective of this study was to investigate the antimicrobial activity of Scabiosa Olivieri on gram-positive, gram-negative bacteria and fungi, as well as to study its anti-inflammatory properties by investigating the factors of human IL-1 and TNF-α, which ultimately led to the introduction of an antimicrobial agent and new anti-inflammatory drugs with a natural and inexpensive source. Materials & Methods: First, the extract of the plant was prepared by maceration. Then, the antimicrobial properties of this extract on E. coli, Staphylococcus aureus, and Candida albicans were investigated by MIC. Also, the effect of this extract on the expression of IL-1 and TNFα in human peripheral blood mononuclear cells was evaluated by ELISA. Results: Scabiosa Olivieri's extract significantly showed anti-inflammatory properties and has antimicrobial activity against Escherichia coli and a mild antimicrobial activity against Staphylococcus aureus. But the antifungal property was not observed. Conclusion: It seems that Scabiosa Olivieri's extract can be used as an anti-inflammatory and antimicrobial agent. However, the effects of its use in the in vivo environment and the chemical analysis of its constituent compounds require further research.
